Reprogramming of microRNA expression via E2F1 downregulation promotes Salmonella infection both in infected and bystander cells

چکیده

Abstract Cells infected with pathogens can contribute to clearing infections by releasing signals that instruct neighbouring cells mount a pro-inflammatory cytokine response, or other mechanisms reduce bystander cells’ susceptibility infection. Here, we show the opposite effect: epithelial Salmonella Typhimurium secrete host factors facilitate infection of cells. We find endoplasmic reticulum stress response is activated in both and cells, this leads activation JNK pathway, downregulation transcription factor E2F1, consequent reprogramming microRNA expression time-dependent manner. These changes are not elicited bacterial pathogens, such as Shigella flexneri Listeria monocytogenes . Remarkably, protein HMGB1 present secretome -infected responsible for IRE1 branch non-infected, Furthermore, E2F1 associated alterations promote replication within prime more efficient

MicroRNA-217 Promotes Angiogenesis of Human Cytomegalovirus-Infected Endothelial Cells through Downregulation of SIRT1 and FOXO3A

Human cytomegalovirus(HCMV) infection has been shown to contribute to vascular disease through the induction of angiogenesis. However, the role of microRNA in angiogenesis induced by HCMV infection remains unclear. The present study was thus designed to explore the potential effect of miR-1217 on angiogenesis and to disclose the underlying mechanism in endothelial cells. We found that HCMV infe...

Salmonella enterica serovar-specific transcriptional reprogramming of infected cells

Despite their high degree of genomic similarity, different Salmonella enterica serovars are often associated with very different clinical presentations. In humans, for example, the typhoidal S. enterica serovar Typhi causes typhoid fever, a life-threatening systemic disease. In contrast, the non-typhoidal S. enterica serovar Typhimurium causes self-limiting gastroenteritis. The molecular bases ...

miR-34a induces the downregulation of both E2F1 and B-Myb oncogenes in leukemic cells.

PURPOSE To elucidate new molecular mechanisms able to downregulate the mRNA levels of key oncogenes, such as B-Myb and E2F1, in a therapeutic perspective. EXPERIMENTAL DESIGN B-Myb and E2F1 mRNA levels were evaluated in primary B chronic lymphocytic leukemia (B-CLL, n = 10) and acute myeloid leukemia (AML, n = 5) patient cells, in a variety of p53(wild-type) and p53(mutated/deleted) leukemic ...

Hepatitis C Virus Infection of Cultured Human Hepatoma Cells Causes Apoptosis and Pyroptosis in Both Infected and Bystander Cells

Individuals infected with hepatitis C virus (HCV) are at high risk of developing progressive liver disease, including cirrhosis and hepatocellular carcinoma (HCC). How HCV infection causes liver destruction has been of significant interest for many years, and apoptosis has been proposed as one operative mechanism. In this study, we employed a tissue culture-adapted strain of HCV (JFH1T) to test...

Reprogramming immune responses via microRNA modulation.

It is becoming increasingly clear that there are unique sets of miRNAs that have distinct governing roles in several aspects of both innate and adaptive immune responses. In addition, new tools allow selective modulation of the expression of individual miRNAs, both in vitro and in vivo. Here, we summarize recent advances in our understanding of how miRNAs drive the activity of immune cells, and...
